Sorting and processing of secretory proteins.

چکیده
a common biosynthetic origin in the rough endoplasmic reticulum (RER), from where they are transported to the Golgi complex. It is in the trans-Golgi network (TGN) that proteins destined for the regulated secretory pathway will be sorted from those to be secreted via the constitutive pathway. Both of these pathways involve vesicular transfer to the plasma membrane followed by the secretory event itself, exocytotic discharge of vesicle contents. In many instances, secretory proteins, even after cleavage of the signal peptide in the lumen of the RER, are in the form of a higher-molecular-mass precursor or proprotein. Conversion of proprotein to protein occurs in both the regulated and the constitutive pathways, but in different compartments and as a result of the action of different endoproteases. Sorting in the TGN and conversion of proproteins will be the major focus of this review.

Biosynthesis and secretion of pituitary hormones: dynamics and regulation.
Production and secretion of hormones by the pituitary involve highly orchestrated intracellular transport and sorting steps. Hormone precursors are routed through a series of compartments before being packaged in secretory granules. These highly dynamic carriers play crucial roles in both prohormone processing and peptide exocytosis. We have employed the ACTH-secreting AtT-20 cell line to study...
متن کاملRegulated Secretion: Helper proteins for neuroendocrine secretion
The granins, known to be general constituents of neuroendocrine secretory granules, are regulators of the proteolytic processing of peptide precursors and promote their aggregation-mediated sorting into secretory granules.
متن کاملProtein Profiling of the Secretome of FcεRI Activated RBL-2H3.1 Cells
Background: Secretory proteins of IgE receptor activated mast cells and basophils play a pivotal role in the generation of immediate and long term immune responses in allergy and type I hypersensitivity. Objective: The present study aims to generate a 2-D map and profile of proteins secreted from a high secretory variant of the rat basophilic leukemia cell line, RBL-2H3.1, which in view of the ...
متن کاملIdentification of excretory-secretory products from larval stages of Ostertagia circumcincta cultured in vitro by SDS-PAGE and immunoblotting
Exsheathing fluid (EF) and excretory-secretory products (ES) of infective third-stage cultured larvae ofOstertagia circumcincta were analysed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDSPAGE). Five and seven predominant proteins were found in the EF and ES products, respectively. Immunoblotting by sheep pre-infection serum did not react with any of the EF and ES proteins, b...
متن کاملN-Terminal Acetylation Inhibits Protein Targeting to the Endoplasmic Reticulum
Amino-terminal acetylation is probably the most common protein modification in eukaryotes with as many as 50%-80% of proteins reportedly altered in this way. Here we report a systematic analysis of the predicted N-terminal processing of cytosolic proteins versus those destined to be sorted to the secretory pathway. While cytosolic proteins were profoundly biased in favour of processing, we foun...
متن کاملEvidence for defects in membrane traffic in Paramecium secretory mutants unable to produce functional storage granules
The ciliated protozoan Paramecium has a regulated secretory system amenable to genetic analysis. The secretory storage granules, known as trichocysts, enclose a crystalline matrix with a genetically determined shape whose biogenesis involves proteolytic maturation of a family of precursor molecules into a heterogeneous set of small acidic polypeptides that crystallize within the maturing vesicl...
